Usually, the lifetime limiting factor for UHPLC columns is high backpressure.
backpressure = 배압(背壓)
=> 흐름 저항 때문에 펌프에 거꾸로 걸리는 압력.
일반적으로 UHPLC 컬럼의 수명을 제한하는 가장 큰 요인은 높은 백프레셔이다.
=> UHPLC 컬럼은 화학적 열화보다 물리적 막힘(압력 상승) 때문에 먼저 죽는 경우가 많다. “컬럼이 오래 못 간다” = 대부분 막혀서 압력 한계 초과라는 뜻.
Particulate matter in the sample is collected on the inlet frit of the column, which causes an increase in the backpressure until the pressure limit of the system is reached.
Particulate [pɑːrˈtɪk.jə.lət] : 입자성의
Particulate matter in the sample : 시료 속에 녹지 않고 남아 있는 고체 미립자
frit [frɪt] : 미세한 구멍이 있는 금속·세라믹 필터
시료 내의 입자성 물질은 컬럼 입구 프릿에 축적되며, 이로 인해 백프레셔가 증가하여 결국 시스템의 압력 한계에 도달하게 된다.
=> 컬럼 맨 앞 프릿(inlet frit) 이 필터처럼 작용하면서
→ 시료 찌꺼기·침전물이 쌓이고
→ 압력이 점점 올라가
→ 장비 pressure limit 에러 또는 컬럼 폐기 발생
Using the inline filter is recommended to prevent blockages of the column frit, especially when sample preparation does not allow for filtering or the sample potentially forms precipitate.
인라인 필터 사용은 컬럼 프릿의 막힘을 방지하기 위해 권장되며, 특히
시료 전처리 과정에서 여과가 불가능하거나 시료가 침전을 형성할 가능성이 있을 때 더욱 권장된다.

These inline filters with a nominal pore size of 0.3 µm protect the UHPLC column from clogging by filtering particulate matter from samples or the UHPLC system.
nominal [nɒm.ɪ.nəl] : 공칭의 : 公(공적일 공) 稱(부를 칭)
=> 이름이나 규격으로 정해진 값
(ex. nominal pore size 0.3 µm : 0.3 µm 기준으로 설계·분류된 필터)
pore [pɔːr] : 공극 : 孔(구멍 공) 隙(틈 극)
=> 필터·프릿(frit)의 입자가 통과하는 구멍
clogging [klɒɡ.ɪŋ] : 막힘
=> 입자나 침전물이 쌓여 흐름이 막히는 현상
공칭 공극 크기 0.3 µm의 이 인라인 필터는 시료 또는 UHPLC 시스템에서 유래한 입자성 물질을 여과하여 컬럼 막힘을 방지한다.
* 0.3 µm인 이유
=> 컬럼 프릿을 막는 입자는 잡되, UHPLC 분리·압력에는 거의 영향을 주지 않는 실무적으로 검증된 최소값
UHPLC 컬럼 inlet frit 의 공극 크기 : 0.2–0.5 µm
인라인 필터는 프릿보다 같거나 약간 큰 입자를 미리 잡아야 함
0.2 µm : 여과 O -> 너무 작아 압력 상승 위험
0.45 µm : 압력 안정 → 보호 성능 부족
Advantages of the inline filter:

*delay volume : 시료가 실제로 컬럼을 빠져나온 뒤, 검출기까지 도달하는 데 필요한 추가로 채워져 있는 부피
-> 분리에 직접 기여하지 않지만, 피크를 늦게·퍼지게 만드는 죽은 부피(dead volume)
=> 인라인 필터를 넣어도 분리에 영향이 거의 없다
2. Specified for working at high pressures (max. operating pressure 1300 bar)
Specified [spes.ɪ.faɪd] : 규정된
고압 조건에서 사용하도록 규정됨 (최대 작동 압력 1300 bar)
Installing the inline filter into the pump is recommended to protect the downstream system from blockages in the following cases:
다음과 같은 경우, 하류 시스템을 막힘으로부터 보호하기 위해 펌프에 인라인 필터를 설치하는 것이 권장된다:
1. When solvent combinations are used that can form precipitates after mixing,
혼합 후 침전을 형성할 수 있는 용매 조합을 사용할 때
2. When running applications with buffers or additives while using columns with small particle sizes
작은 입자 크기의 컬럼을 사용하면서 버퍼 또는 첨가제를 사용하는 분석을 수행할 때
General hints for effective use of the inline filter:
인라인 필터를 효과적으로 사용하기 위한 일반적인 팁:
1. Filter solvents before use.
사용 전에 용매를 여과한다.
2. Follow best practices.
3. For G4204A, G7104A/C, and G7131A/C pumps, backflush the filter in the pump weekly (start Flush Filter from the context menu).
G4204A, G7104A/C, G7131A/C 펌프의 경우, 펌프에 장착된 필터를 주 1회 역세척(backflush)한다
(우클릭 메뉴에서 Flush Filter 실행).
CAUTION
Damage to the valve
밸브 손상
Use the filter flush mode only if the inline filter is installed, otherwise the pressure pulse might damage the multipurpose valve.
pulse [pʌls] : 순간적인 파동
pressure pulse : 지속 압력 X, 순간적인 압력 스파이크
인라인 필터가 설치된 경우에만 필터 플러시 모드를 사용해야 하며, 그렇지 않으면 압력 펄스로 인해 다목적 밸브가 손상될 수 있다.
=> Flush Filter 모드는 ‘필터가 있다는 전제’로 순간적인 고압 펄스를 발생시키는 기능이기 때문에, 필터가 없으면 그 충격이 그대로 밸브에 전달되어 손상을 일으킬 수 있다.
NOTE
For inline filters installed on the sampler, exchange the filter frit (Frit 0.3 µm for inline filter, 5/pk (5023-0271) or Frit 0.3 µm for inline filter, biocompatible, 5/pk (5320-0022)) every 1000 injections or when the back-pressure rises by 15 %.
샘플러에 설치된 인라인 필터의 경우, 필터 프릿
(인라인 필터용 프릿 0.3 µm, 5개/팩 (5023-0271) 또는
인라인 필터용 바이오컴패터블 프릿 0.3 µm, 5개/팩 (5320-0022))을
1000회 주입마다 또는 백프레셔가 15% 상승했을 때 교체한다.
